No, do not spray anything in the throttle body. If the unit is not starting correctly, fix THAT issue. You do not specify anything about your vehicle and model, so little help can be offered. BUT, Gas or diesel, you should not have to spray anything to help in starting, something else is wrong, find and fix that.
Yes - but use sparingly on gasoline engines - it can cause detonation if too much is used. Good idea not to use it if possible on a gasoline engine. It is somewhat safer to use on diesel engines in cold weather since they fire on compression anyway, but it should never be used on a diesel engine at the same time glow plugs are being used.
Some manufacturers of heavy equipment that run diesel engines actually have an ether injection system built into the starting system to aid in starting in temperatures as low as minus 50 degrees F ( arctic conditions - ice road building etc,) Also see Wikipedia - staring fluid - article for more info on starting gasoline engines using ether.
