Another thing that can cause this is the carb. It may be getting too much fuel once it's warmed up. An engine needs more fuel when it's cold than it does when it's warm. Cold engines run inefficiently so they need more raw fuel to fire-up, and stay running. This is all a choke does. A choke plate closes-off the air going into the carb which makes the fuel mixture really rich so it can run. Less air = more fuel. You'll also notice that cars with chokes also idle real high until they warm-up. This fast idle and the over rich mixture (due to lack of air), allows the engine to fire up and stay running. As the engine warms-up and starts burning fuel more efficiently, the choke plate begins to open, which slowly lets more air in and leans-out the air/fuel mixture. Once the engine is warmed-up, the choke plate will be wide open allowing the carb to run as it is supposed to with the correct metering of fuel and air.

#1 and #4 are not firing I changed the coil pack and still no change any ideas

You said you changed the coil pack and the wires but didn't mention the plugs. Try moving one of the plugs to a different cylinder and see if the miss follows it. It could also be the ignition module. (the thing the coil packs mount to.)

Low fuel pressure.
You need to check the fuel pressure on the first crank and watch it as it is running.
Requires a special gauge.
If it were just a starting problem I would say it could be a pump relay, but not if it runs rough for 5 min.
Might be a dirty filter or problem in the tank.

What causes misfire in #1 cylinder

A failed fuel injector will do it also. Compare the injector coil resistance with that of another injector. If it's open or significantly different, the injector coil is bad. The injector could also have a mechanical problem such as a clog.

Seems that the fault relates to heating, check if the fuel pressure is dropping when the engine heats up, allow the car to idle so as to confirm . Also this will indicate if there is failure of the ignition system, This can fault in the ignition , failure of the HT coils.
What is important is your observation to the case before any part is replaced.

First you need to disconnect each spark plug wire while running and find out which cylinder is unaffected by this, or get the codes read from the check engine light at an autozone, to figure out what cylinder is misfiring. If you don't gave any codes present have the fuel pressure checked and go from there

im a tech in the bronx and i dealt with the same code for the misfire. burt i realized that there wasnt any power to the coil of #2 cylinder. I replaced the sensor mounted on top of the air filter box, which solved this problem.

2000 Pontiac Grand AM trouble code P0300

Im surprised that didn't fix it. On the Quad4 engine its usually a cracked coil housing.
Have you checked:
  • fuel injectors / fuel pressure
  • coolant temp sensor
  • compression - to check for bad head gasket
  • ignition control module
  • EGR system - make sure the valve isn't stuck
  • crank position sensor.
The crankshaft position system variation compensating values are stored in the PCM non-volatile memory after a learn procedure has been performed. If the actual crankshaft position system variation is not within the crankshaft position system variation compensating values stored in the PCM, DTC P0300 may set.
