It will UNLOCK remotely if locked manually. The UNLOCKED warning light shows. A quiet beeping is heard from the back left corner of the engine bay for about 10secs. on attempts to lock. All doors bonnet and boot lid appear to be secure.
Be sure you don't have a spare fob in the trunk. Cars will not let you lock with the "keys"in the car. My wife would try to lock her purse in the trunk to avoid carrying it and we had the same thing.
Get a readout from the ODB bus to see which, but it could be: -The transistors (solid state relays maybe) for locking are ready to replace. -Cable breach or waterlogging somewhere from that lock module (that also makes noise) to the door solenoid bits. Salted up connector at firewall, maybe? -Time to lube the lock; it's having to push hard to lock it, but unlock has a priority because you want to be able to escape your car. -Just a bad or worn door switch, claiming overtight closure. (If the door were just open, that beep doesn't happen, right?) -Door seal actually out of spec (collapsed, say,) keeping the lock from lining up right.

I have found a ridiculous solution for this door lock issue. As many people have mentioned, it does involve the control panel under the driver side dash. I noticed that if I pushed the control panel/fusebox in while working the key fob and/or door lock switch, IT WORKED! So, my silly solution was to insert a t-ball (regulation size) between the fusebox/control panel and the paneling closest to the driver on the top side of the fusebox. IT WORKS!!
Glad I didn't spend $500 for Toyota dealer to fix.
Hope my son doesn't miss his t-ball!!

the actuator might have been disconnected from the lock, its attached inside the door, with a metal rod that clips into the actuator, check it out, might be a easy fix, good luck and god bless, please rate me!

If your car has an alarm with a separate remote, it is a separate system and it is probably self-arming. When you lock the doors with the key central lock, the alarm gets armed by the RAV4 being OFF and the door locking action. When you unlock the doors with the key, the alarm is still armed because the key is not comunicating with the alarm just the cars factory keyless system.
Remote Control Fob Procedures:
The Central Door locking is controlled by the Multi-Remote Control system which controls
the door locking, door super locking and emergency hazard reminders.
When the doors are locked and unlocked the unit flashes the hazard lights once for lock and twice for unlock.
A maximum of four remote controls are allowed to be programmed:
ALMERA
ALMERA TINO
PRIMERA
MAXIMA
MICRA
SERENA
VANETTE CARGO
TERANO II
Ensure all the doors have been unlocked, either by using a good transponder key or remote
control plip key.
Procedure:
1. Turn the ignition switch from Position 0 to ON 6 times within 10 seconds.
2. Next, turn ignition switch to OFF position. Leave key in ignition switch.
3. Wait 2 seconds then the system will enter programming mode and will flash the warning
lights twice.
4. Press and HOLD the unlock button on the Plip.
5. While pressing the unlock button, press the lock button 3 times.
6. Release the unlock button.
7. The warning lights will flash once to indicate successful programming.
8. Repeat procedure 4 to 7 for up to 4 plip key's.
9. When completed turn ignition ON, and the warning lights will flash 2 times.
10. Remove key, and check all plip key's for operation.
NOTE : The programming mode will stop when either the ignition is switch ON, 4 plip keys
have been programmed or no input signal either from the switch or plip keys has been received
for 120 seconds.
