SOURCE: Wrong Way
Mine does the same thing, I think it's because of the settings to "find fastest route". I just ignore what it says and go the way I choose, it recalculates the route and all is well.

Latitude is N or S, longitude is E or W, correct.
When entering degrees, minutes and seconds, you dont have to type the double apostrophe at the end off seconds, the device will understand...
ALT and LONG can also be seen as decimal degrees,
as in your case N48.87329 E8.6923
The TT device understands them both, just type them in....
SOURCE: TOMTOM ONE MAP GREECE
There is in face a map for Greece. Tele Atlas provides mapping solutions for TomTom products, and they do have maps for Greece for the TomTom ONE 3rd Edition. Select the device from the pull down menu on the Map Store, and you will see there are indeed updated maps for Greece.

You were able to successfully download windows files with your Mac and transfer them to your Tom Tom? I'd suggest trying the transaction again, but doing it with a windows machine. (Trust me, I have a Mac and I realize how unwell it likes to play with Windows.) {But I wouldn't trade it}
