I have a new Samsung Printer (SL-M2820DW). I want to print index cards, ideally duplex. But when I fill the tray (not the manual feeder), I get an error message saying that there is no paper in the tray. What am I doing wrong?

Menu> scroll to Paperhandling> MP Tray type>>(plain) try changing this to cardstock <enter>
instead of MP Tray type do the same for Cassette if you desire....

SOURCE: The printer will not pull card stock paper
Usually the stock is too thick to process properly.
Go to a lighter stock or buy a more robust printer if you are going to do a lot of that type of work

"MP" tray is refering to the top fold down tray on the front of printer (bypass tray) You might load paper in the bypass and see what it does?

check the paper type and the paper size from the printer menu, if it is ok then you will try to check the paper sensor on tray 2
