When using a 12v inflator for tires, keep the car running or not?

Yes, you should keep the car running, but you don't have to. If you decide not to, then you can drain your car battery, and then the car won't start.

Looks like you overloaded and blew the cigarette lighter fuse while using a 12v tire inflator pump. Check the fuse and replace it.
