I have dust particles inside
No. There is no easy way to remove them, to do so requires an expensive strip down and overhaul in a clean dust-free environment.
Unless you have the dust equivalent of boulders in there, then they are highly unlikely to show on your images and you can ignore them. Even if they do show occasionally you can usually easily correct the faults using commonly available processing software.

Well, that's a great lens and most definitely worth the cleaning/service. I don't know where in the world you are but fungus growing inside a lens is only heard of in high humidity environments and can happen fairly quickly. Are you sure this is a fungus on the inside and not just a clouded smear on the front element? If the lens is having a problem then the camera won't be to far behind. To answer your question more directly "what should I do" my suggestion is to send the lens to a authorized Tamron service/repair center/depot for cleaning. After which get a few of those little moisture absorbing packets and place them in with your photo gear to help reduce the moisture.
Auto focus not working
Sorry but if you've tried all the usual fixes then there's nothing more that you can do. Either pay for a professional repair or continue to use the lens manually.
A professional repairer should be able to indicate what the likely costs are going to be and you can then judge whether repairs are cost effective.
Personally, I'd stick to manual focus as I've always preferred knowing exactly what I'm focussing on instead of constantly fiddling with buttons and menus to override what the dumb camera has locked onto.
