After driving on the highway for a distance and getting off at an exit or stopping for any reason there is a knocking sound and engine appears to be missing. This doesn't happen with town driving or driving under 55 mph only after driving on interstate. Problem just started about a week ago but it appears to be getting worse.

Check that the oil level is correct as extra oil foams up and the oil pressure falls. Around town driving does not put a strain on the cooling system but continuous highway driving does and shows up as blocked cores and collapsing bottom radiator hoses. Ignition timing is critical at high speeds so ensure that that is correct. Check that the temperature gauge is accurate as the noise and missing is indicative of an overheating engine.

You need to have it scanned. The od off light is like the check engine light for the transmission. It will flash when you have transmission codes.

What could be causing a knock from the rear of the car on the highway when not accelerating?

Best guess would be a universal joint going out on your rear drive line. Check that out real soon because the could be dangerous if that goes out on the highway!

Sounds like you might be burning up your ATF on a long trip. Try installing a transmission cooler and add some lucas oil transmission additive.

Check to see if your ATF smells burned.

Toyota solara engine knocking and car cutting off

Meloe77, there's a high possibility you have a mechanical failure inside the engie causing the knocking sound. (Check the oil?)
The reason for shutting off is because the knock sensor can hear it, and the powertrain control module (PCM, herefore) is trying to detune the engine to eliminate the sound, (as if it were caused by bad gasoline.) The fact that there is a mechanical knocking sound, means that the sound cannot be eliminated by detuning of fuel or spark mapping, therefore it starves for fuel and/or spark causing the engine to stop running.
My recommendation: Have the vehicle TOWED to a reputable mechanic and ask him if its possible that I'm right.

How safe is it to drive long distance with the engine knocking

No additives are ever needed in any vehicle,
nor will they fix any mechanical problems

I would stop using it & have the issues, at
least diagnosed, to get a basis of where your at

My check engine light is on and when it is on my transmission seems not to go into gear right and my car is stsarting to knock my dad said it could be the lifter but not sure

If you hear a knock, generally it is in the rotating assembly. (wrist pin to the piston, or main
bearings) If it is more like a clicking sound then it could be a lifter the sound will generally
be confined to the upper portion of the engine
Usually if an engine is failing it will exhibit poor power which may seem like
transmission problems.

(I experienced this first hand driving a 66 GTO on a long distance trip, the knocking
became louder and then two rods exited the engine through the oil pan).

1997 LeSabre has engine "knock"

If it knocks at 50 it will knock sitting still if you race it up. Is the oil pressure good? These motors hardly ever have a bearing problem. The only knock problem was the harmonic balacer that comes apart and knocks. The rubber falls apart and it will make a knock sound all the time, more with the ac on. If you have a knock at 50 or highway speed only you may need a outer tie rod end only. Get the front end checked as they will make a knocking sound on the highway. reply
