Your steering locks could be in a bind. Is your steering wheel turned to the side? Try moving the steering wheel while trying to turn the key. If this is not the case, then you most likely have either a worn-out key or a worn-out ignition lock cylinder assembly.
I would first try a different key if you have one. If there is not another key, I would take the Vehicle Identification Number (VIN) to the dealer along with proof of registration and picture ID, and have them cut a NEW key using the key codes for your vehicle. If this does not work, then you will probably have to replace the ignition lock cylinder assembly.
This typically means your park/neutral switch is failing since it works after moving gear selector, here are instructions to replace.
DISCONNECT OIL COOLER PIPE
REMOVE PARK/NEUTRAL POSITION SWITCH
Disconnect the connector.
Pry off the lock washer and remove the nut.
Remove the bolt and park/neutral position switch.
INSTALL PARK/NEUTRAL POSITION SWITCH
Install the park/neutral position switch and bolt. Torque: 13 Nm (130 kgf-cm, 9 ft. lbs.)
Install a new lock plate and the nut. Torque: 3.9 Nm (40 kgf-cm, 35 inch lbs.)
Bend claws on the lock plate to fix the nut.
Connect the connector.
Check that the engine can be started with the shift lever only in the N or P position, but not in other positions. If not as stated above, carry out the adjustment procedure.
CONNECT OIL COOLER PIPE
FILL ATF AND CHECK FLUID LEVEL
TEST DRIVE VEHICLE
The problem has something to do with the anti-theft module under the hood in the black box near the battery. I have talked with toyota factory, toyota dealers etc. They are no help and want to replace the entire ignition. But when I am stranded, I raise the hood and tap on all the modules ( since I cannot determine which) and the key will turn and start the engine. Solves problem for 3 or 4 weeks. Then same **** thing all over again. This problem makes me madder than the rear door latch breaking over and over again. Toyota getting more like the US manufacturers with their KMA attitudes at times.

The steering locks don't necessarily engage when the key is removed, I always turn the wheel before exiting to engage it. There are several positions that will allow the pin to lock in but if between these, it may seem that it isn't locked.

the steering wheel SHOULD be locked if the key is not in the ignition and turned to the 1st position. If you can turn the steering wheel more than an inch or so without the key n the first position, the ignition switch (read: thingy you put the key in) is bad.
Sometimes you may have to move the steering wheel slightly to one side or the other to get the key to turn, this in itself is not a cause for alarm. Try it and let us know. If this doesn't work, you are probably looking at a couple hundred dollars parts and labor to get the switch/steering wheel lock mechanism fixed.

Hi what is happening is that your turn off your car and left the wheels turned to one side and specially if your car is parke in a hill what you have to do is to force the steering wheel to the same direction of your wheels this will free up the ignition switch it is going to be hard let me tell you but it will work and this is why your shift don't work once you have the ignition switch free then your shift will work again

is it correct key? key and ignition barrel could be worn wiggle key around in ignition while turning? gently but fast wiggling.try lube in key slot also.

I have a 2001 C240 with the same issue. I removed the positive battery cable from the battery and kept it off for 30 seconds then replaced it. Everything was reset and the key worked.
