If you are talking about the High Brake Light at the top of the tailgate:
1.) Take out the two Phillips head screws in the lens.
2.) Carefully pry the lens straight back . The gasket sticks to the paint so be careful not to rip the foam gasket.
3.) The bulb is a 921 replacement. Easily found size.
If you're talking about the brake light in the tail light assembly:
The instructions for accessing the tail light are in the Owners Manual. Basically, you have to remove one or two screw/bolts from the inside of the vehicle, then there are two snap type plastic hole/receptacle in the plastic tail light assembly. Be very careful that you pry on the housing in the proper direction, perpendicular to the entry of the snap in connector, or you will break the plastic bayonet or attachment to the lens assembly.
It sounds harder than it really is once you take a look at it. If you have a manual, it will tell you and show you how to get the rear tail lights off easily without breaking any of the plastic connectors. Hope this helps.

I have a 2001 Sienna and have had to replace most of my tail light bulbs.
The bulbs that are mounted in the fender (wrap arounds) have acess panels in the van's interior that easily pop out and expose the back of the light assembly.
However, the bulbs that are found in the lift gate are a different matter. You have to remove the whole interior panel to the lift gate to expose the light assemblies. Go to your auto parts store and get a tool to help pop out the plastic retainers from the lift gates metal frame. Don't force the retainers out. Go slowly and gently so that you don't damage the interior panel. If you are really gentle you might even get to reuse the retainers.

No special tools at all. Are you familiar with doing brakes? I wouldnt recommend it for a first timer, its not worth the lives of you and others. Ive also seen people take the wrong bolt out of calipers and spend triple what a brake job cost because they werent experienced and wanted to save a buck. A 17mm and pad spreader or big channel locks is all you need. Use Toyota pads and have the rotors cut..OR... Dont complain when they make noise all the darn time!
