have a windscreen shop check for proper sealant under screen
( may have to remove the screen and check for rust around the glass or have new sealant fitted properly)
usually in pairs two of one type threaded into exhaust before catylitic converter and two of another type threaded into exhaust behind catylitic converter(looks like little muffler in exhaust close to engine.)

the coolant needs to be serviced @ 60k according to lexus reccomendation. theres a timing belt that needs to be changed @ 90k miles. I usually just change the coolant with the timing belt because you have to take the water pump off anyway and replace the engine coolant.
no there is not really a downside dealers are good it doesn't really matter who does it but lexus is more familiar with lexus vehicles than toyota because they specialize in it but something like that is simple .
U do not have to use synthetic oil but I recommend u do if u plan to drive the car for 100,000 miles or more, syn oil almost eliminates engine wear, if u use it change the oil and filter every 5,000 miles
