I don't think you can assess the condition of the transaxle by spinning the wheels. The differential in the transaxle converts power from the transmission to the wheels by a set of gears. It would almost impossible for one side to turn at a different speed than the other. You may have more drag on the brakes on one wheel, or the fluid in the transaxle could cause one side to go slower with the van off the ground.
It sounds like you have a bad spindle/hub. You will need to jack up the front left tire, remove the wheel and the brake caliper and pads. Now spin the hub by hand. If you hear a grinding like sounds or low rumble sound, or there is more resistance than normal, replace your hub/spindle assembly.
Could be road noise from a shifted belt in the tire. could be a drive axle going out. those wheel bearings dont hum 99% of the time when they go bad,generally they have a low grinding sound,espesially when you turn,and your teering gets a bit loose and sloppy.
also check to make sure that you dont have a broken strut that may be touching the tire..try putting on your spare and see if that fixex it,if so its a shifted belt in that tire.
Jack up the wheel that may have the bad wheel bearing and position your left or right hand at the top of the wheel and the other hand at the bottom of the wheel and rock from top to bottom if there is over 1 cm of play your wheel bearing needs to be replaced and if there is excesive play from left to right your rack ends need to be gresed if the have a grease nipple or replaced hope i was some help
