Have you checked or have had the battery tested? If you need to manually lock your car, most cars that have a fob, also have a key built in to lock/unlock your vehicle.

There is only one fuse that mentions 'Locking', but also refers to the General Electrical Module (GEM). If you change the fob battery, you need to reprogram it to the vehicle. Or you could open any Transit around you. Lots of help below including resetting..
Because you removed the fob battery and further compounded by the fact you disconnected the vehicle battery, the complete lock system and the anti-theft immobilizer system needs reprogramming. I am undecided whether a locksmith or an electrician is better, but would suspect the auto locksmith.
The car should always be started with a proper chipped key. The valet key on my car has a chip in its credit card style case. It will start the car if the case is close, or you can give the key to someone just to gain entry to the car and not start it.

We just spent $1000 Canadian at the Buick dealer having trouble shooting and repairs done on our 2006 Terraza.
One of the remotes had an internal broken circuit.
Had to replace the Body Control Module on the AZTEK.
Kissed another $1,000 goodbye.
Selling it.
Frankly, as long as they are working it's fine, but when they start to have problems, it is completely beyond the scope of THIS Fixya ****** to diagnose or attempt to repair.
Makes me want to sell and NEVER get another GM.

Check the light on the remote fob. Does it light up when you press the button? It will light for only a part of a second.
Sometimes the transmitter stops working for no reason and there is no repair, you have to buy a new remote fob, or in the case of some vehicles, you have to buy a new key and have the agents activate the key, then you can activate the remote with a special procedure.
Be sure to use the correct battery, properly in place, clean the key contacts , See owner's manual for additional information.
There are specialized locksmiths for car keys
