The code should not have any effect on if the engine runs or not.
Code 500 is for a problem with the speed sensor in the transmission.
You'll need to find out if you are loosing fuel or spark when the engine stops.

Transmission speed sensor

You could have a faulty speedometer/odometer combination meter causing a false Vehicle Speed Sensor error.
My 1998 Sienna had the following symptoms:
- Speedometer and odometer failed intermittently
- The check engine light would come on and I would get a p0500 code in the OBDII.
- The cruise control would stop working at the same time the speedometer failed.
There is a speed sensor in each one of the tires and they feed the speed information to the ABS ECU. From the ABS ECU the speed signal goes to the speedometer/odometer combination meter, and from there it goes to the Engine Control Module and to the Cruise Control ECU. I could see a signal coming into the speedometer/odometer, but no signal going out. Replacing the speedometer/odometer combination meter fixed the problem.

I had a P0500 code on my 2006 Toyota Sienna AWD limited edition. I changed the speed sensor and now it will not change gears. Does the transmission need to be reset or is there another issue I am missing?

Where is the speed sensor on a 1998 toyota sienna

The 1998 Sienna has one speed sensor in each tire (ABS sensors.) These sensors are used by the ABS system, but also to calculate the vehicle speed and for the tire air pressure monitor. If you are getting a P0500 code and your speedometer does not work, your problem could be a faulty speedometer. If one of the speed sensors were bad, the low tire air pressure light would come on. this obd2 trouble code table can help you understand the meaning of code.
For this code you can try this:
The simplest thing to do is to reset the code and see if it comes back.
for your car, I think the ps701 jp scanner can reset the code.
Next, check all wiring and connectors that lead to the speed sensor. The location of the sensor depends on your vehicle. The sensor could be on the rear axle, transmission, or perhaps the wheel hub (brake) assembly.
If the wiring and connectors are OK, then check the voltage at the speed sensor. Again, the exact procedure wil depend on your make and model of vehicle.
If everything looks OK, replace the sensor.
