South African built 2.0 litre Golf 5. When inserting the MP3 recorded disc it ejects with an "Error" message. It plays in other cars with MP3 capability.
If the radio was not originally built to play MP3 format, then no. Unless the manufacturer has an update for the player's software/firmware (which I doubt they bother with) it will be impossible.
Try inserting another mp3 disc in the player. if it ejects that one as well, your player cannot play the format.

The question was whether the player can be set to play MP3.
if it doesn't play them then it's unlikely it can be set to play them without updating the radio firmware which you would need to talk to VW about (or the radio manufacturer).
