All started when the cheak engine light came on for transmission component slipping

check the brake light circuit (do the brake lights work), this is what provides power to unlock the shifter, it is called the brake park interlock and is one of the new DOT safety devices..

it's possible that the brake light switch on your brake pedal arm could be bad. this controls both the brake lights and the solenoid that unlocks the shifter to allow the button to be depressed. as far as the truck not starting, the shifter linkage may need adjusting on the transmission.

P0621 - indicates a failure in the alternator lamp curcuit.
P0304 - indicates a miss fire was detected in the #4 cylinder
U1040 - indicates a loss of communication with the ABS system
U1000 - Indicates a class 2 communication loss
None of those codes are relating to a transmission failure (P0700-P0702)
I would have a visual look at the transmission and verify that all electrical wires are connected and tight. Then I would have the transmission serviced if it hasn't been within the last 35,000 miles.
Have the trouble codes cleared and address the ones that return.
After that you may be looking at an internal transmission adjustment and/or repair.
Good luck.

Check both O2 sensor(s) and catalytic coverter.. P0420 means catalyst below threshold. Also check the sensor at the transmission and solenoid.

Bring to tranny shop or repair shop, have it scanned for trouble codes, many electrical parts and solonoids in those trannys, possible it may just be a shift solonoid bad or needs adjustment, much cheaper than replacing tranny. good luck!
