You will have to take the door panels off of each of the doors and see what is preventing the latch to lock the doors, whether its just missing the lock or something is broken in the lock mechanism

This sounds like you may have a bad lock actuator, there is an actuator that controls the lock, it may need replacing, you will need to remove the door panel to gain access to it, then with a testlight you will test to make sure the actuator is getting power and a ground and if it tests good for power and ground then replace it and door lock will start working again, this part is about 30.00 dollars. here is a picture of it. good day
Sounds like the drivers door lock switch is bad an as far as the auto start remote the contacts under the button get dirty and won't make contact, open the remote like changing the battery and you can probably get at the contact button, if it's a little carbon button they can be cleaned with a pencil eraser and then paper tower with a bit of rubbing alcohol, if it's an actual switch you can try spraying some contact cleaner into the switch. let it dry before you put the battery back in.
U will have to take the inside door cover off and manualy open it. And replace the electric door locking switch. Very common problem. Get the new lock and latch assembly from dealer,
The title suggest the door won't unlock unless the remote is used but during the summer it was the inverse. If while inside the car the doors will not lock or unlock from the door but will from the remote then you have a faulty switch.If while outside the car the key turns while in the lock but the door won't lock or unlock then the linkage at the end of the lock is faulty. You'll need to remove the door inside plate to check this.
Many Chrysler vehicles won't let you lock your keys in. If locked from the inside, or locked before closed, the doors unlock upon closing. If you don't have a remote, you'll have to lock the doors with the key after you close them.
