To test a circuit breaker you should set the multimeter to "Volts AC" and touch the prongs to the breaker's terminal screw and a ground. If there is no power, the breaker is faulty and requires replacement.

the first thing i would do is to remove the feed wire from the breaker and see if will reset if it does you have a short in the system , tell me more about the system and maybe i can advise further

Yes the maximum size branch circuit breaker that will fit in a residential load center is 100 amps. I would recommend that you calculate the amount of power that you need the sub panel to handle, then add any additional loads you may need in the future. Hopefully this is less than 100 amps. I would recommend that you check with your local electrical inspector to make sure your installation meets all local and national codes before proceeding with the install.
Two circuits in panel inoperative
not likely check the neutrals on those circuits the chance that 7&19 are bad and everthing in between is good is almost impossible. i would check the wires very close and also inspect the neutral buss connections both main connections and circuit connections
