Well, there are many possible reasons that may cause this kind of "unable to access" error with a removable media. But most cases the memory stick must be corrupted or crashed just because of Time (you know it is not life-long guy). In this case recommend you take this recovery program FileRetrieval and see if you can recover the data as much as possible. Then format the memory stick. It should work normally again. It's not over, re-use the program to see if you have more harvest. Best regards.
First if possible try it on another computer, if it doesn't work on it then it's dead (However in my 15 years of I.T. i haven't seen one actually die yet) If it works on the other computer then you will just need to reinstall the usb software on your other computer.
Also if you have to many USB Peripherals in use then they may be hogging the power that your usb device needs to start
