The screen that does NOT flash will need replacing.
It may look fine and dandy but there is still an issue.
The reason for the flash is that when switched on the DS lite performs a qucik check of the screens, if there is an issue the unit will switch itself off again.
My answer is, judging that you have said you have taken it apart to have a look that you are willing to repair it yourself. Have a look on ebay for the relevant screen (plenty of people on ebay stock them) and then replace the screen that does NOT flash.
This should solve your issue.

try getting an R4DS card and download games from www.filestube.com if you dont want to repair it.
you can have 50+ games on an sd card and not have to lug around all those games anymore

Using One Card to Host the Game
Step 1
Look on the game’s
packaging for the "Multiplayer" icon. If the game requires only one
player to have a copy of the game, the icon will show two DS Lites: a
DS Lite with a black screen transmitting waves to a DS Lite with a
white screen. Below the two DS Lites is a range of numbers which
indicates how many players the game can support.
Step 2
Put the game card in the Nintendo DS Lite that is acting as the host for the multiplayer game session and start it up.
Step 3
Have
the other players turn on their systems. They will use the main menu
screen to access the "DS Download Play" icon. They need to tap the icon
to begin downloading the game information from the host DS Lite.
Step 4
Begin playing when all of the systems have finished downloading the game information from the host.
Multiple Cards for Multiple Players
Step 1
Look
at the "Multiplayer" icon. If every player in the game session needs a
copy of the game in order to play, the icon will show two DS Lites,
both with black screens, transmitting waves toward each other. A range
of numbers is under this graphic and indicates how many people can play
at one time.
Step 2
Read
the game's instruction manual on how to set up the game session. The
steps required vary by company and sometimes by title within the same
company.
Hi. this issue is normaly caused by bad screen (normaly top screen) or bad assembly in one of two screens, re insert carefully the flex cable again, if your screens are fine this must fix the issue
the r4ds has a reset application when you press random buttons quickly and the r4 turns of sorry but i think you can turn it off by pressing the R BUTTON on the game loading screen... please pm me for any more help...

and as far as a defect....anything man made ushally breaks at some point...unless its cast iron and 600lbs and built to with stand a war.
You turn de DS Lite on, the green light flashes and the unit turned of. Is that correct?
The DS Lite checks some things before he starts-up. One of these things are the screens. the DS sends a signal to it and expect an answer back. When this answer is not received, it shuts down. The most common problem in these cases is the flatcable of the upper screen. Question is, did you replaced the housing? If so, you might dammaged the flatcable of the upper screen. If not, the flatcable might be defective because of using the DS Lite a lot (open /close the unit).
So first check the flatcable of the upper screen (google and you will find manuals how to open the DS Lite but be carefull!!!!)
Second, on the mainboard are two fuses (F1 and F2) . Check these! One of them might be defective
You may have a bent pin on the game slot. Try inserting a game several times rapidly in and out. Look at your game cartridges and see if one of the contacts is damaged in the same place on all of the cartridges.
If this does not solve the issue, you will need to get your console repaired. Either the repair shop will replace all of the electronics or resolder a new game slot on your existing board.
