Sounds like the spool and/or the spring is bound up by the cover. Try loosening the screws a little at a time and see at what point it returns. Also check under the spool and see if a wood chip may be under the spool that is causing it to bind.
did you change the Spool assembly from the original? The Straight shaft trimmers turn counterclockwise and the curved shaft trimmers turn clockwise. If you put a Straight shaft spool on a curved shaft machine, it will unscrew itself. and Vise verse.

If this is a fishing reel, pull off the old line into a plastic bag or wind it on to an old spool so it can be safely discarded. Tie the new line on to the reel, and wind the new line on.
There used to be a closed face spinning reel that you would unscrew the face cover to access the spool.
If this is a weed eater, grass whip, some of the older units had a threaded centre with a left hand thread. Once unscrewed, the spool lifted off and you could wind on the new cutting cord.
Unscrew the head's center bump-knob counterclockwise with your fingers.
Pull the bump knob off and take the spool cover off the head. Slide out
the spool and any remaining cutting line. Lift out the spring underneath
the spool. Take out the slide washer. Unscrew the arbor bolt either clockwise or counterclockwise, depending
on model, in the center of the trimmer head using the wrench. Take the
entire head off the end of the shaft.
According to the intructions and I quote "Take off the cowling that covers the spool, twist off the pick up mechanism clockwise and a half turn anti clockwise will remove the spool"
