Fault to the main power supply regulator section. Either it will be a separate board, or the part of main board. Check and replace damaged component/s at its main power regulator [SMPS] section circuit. Contact any service technician. If you wish to get some details; check the site linked here. Pull up older posts.

It will be best to replace the power supply regulator board as card basis rather than making "Surgery" to it; which usually will make loss for both money and time. Even though you can make it right, it will cost around an amount equal to the cost of a new board; including the cost of (spare parts + service charge) More to say, most of the spare parts to it will be difficult to get from open electronic spare part market. This is true to most of the flat panel TV power supply boards; irrespective of its brand and screen size.

Make sure you don't accidently have it muted on the remote, and that the volume is up. Otherwise, you'll need to go into the menu and make sure the TV speakers are on, and NOT the remote speakers.

This article is in Spanish but can help you solve the audio problem in your TV.
When the IC2 circuit does not work properly, the failure of the audio malfunction may occur. Trade to update the firmware of the TV over the network or with the reprogramming of the TV EEPROM.
The article is a guide for when the TV only changes channel but for the volume that does not respond can be the same cause.

There is no such thing as a defuse board in a LCD TV, there are defuse screens in the panel is that what your talking about?

Why does message come up no media device

Look in the manual that came with the TV.
It will tell you that you are trying to get a signal from a device that is NOT hooked up to your TV. You probably need to go to 'Input' with your remote, and work it so the 'Input' shows Cable, or Satellite, or DVR or whatever sends the signal to your TV.
The TV is smart enough to know there is no signal coming from the currently selected input.
