Online virus removal can be a bit tricky at best - often trading their 'free' service for (other apps with) malware, spyware and other viruses. A better option might be to install a free antivirus program instead. AVG Free and Windows Defender/Microsoft Security Essentials (for Windows-based PC's) are both great options - among several others, and won't cost you a cent.
The only thing I can think your problem may be is something in "mouse settings" is wrong, try looking. You may still have a virus--go online to Kaspersky or bit-defender for a free virus removal tool, double check. Did you get the virus using IE8? try using Chrome or Firefox. Microsoft Security Essentials is no longer supported for XP, download a free Antivirus such as Bit-Defender (the best right now).

Make sure you don't have any other virus program running such as Norton, Mcafee, AVG, etc. If you do then that will stop windows defender from turning on. If you do have any virus programs running you'll have to uninstall them before turning on Windows Defender.

Click to "remove" it
Restart your computer
Download from http://www.microsoft.com/antispyware
and install the latest version, and let it update.
If it still happens, you may have a computer-virus that is "disabling" Windows Defender everytime you restart.
I did a quick search for 'defender pro 2008' and saw tons upon tons of complaints of it causing many problems. I would uninstall the software and use something else. A program with so many bugs will probably do more damage if you install and run it.
For anti-virus I would suggest AVG Free, as it is free and works better then those big name brands with their bloated software. Your windows firewall should be good enough for the basic PC user. To remove spyware/malware check out another free program called SpyBot Search & Destroy.
To remove Defender, go to your Windows Control panel and click on Add/Remove programs.
There seems to a problem with your win defender turn off defender and try to install the anti virus software as when you have one anti virus or a firewall running it will not allow another anti virus to be installed. Also check if you have any other anti virus installed
