Unfortunately, you are missing some software. You need the original PE-Design software CD. Then you can install the Upgrade software from https://support.brother.com/g/b/downloadtop.aspx?c=us&lang=en&prod=hf_inov4000deus for your OS version. Until this software is installed, the computer will not find the driver for the sewing machine. The driver is not available from Windows Update.
The USB drive may have new firmware that wasn't properly installed. To load firmware, insert the USB drive when the sewing machine is off. Then press and hold the Automatic Threading button while you turn on the sewing machine. Follow the directions on the screen (see the manual on page 224: https://download.brother.com/welcome/doch000013/nv4000ug04en.pdf ) . Turn off the sewing machine and then remove the USB drive before sewing.

Most memory sticks will work, but the smaller capacity sticks work the best.
If your stick has a lot of designs on it, the machine must read all the designs before bringing up any on the screen. Only keep a few on a tick at a time.
The thread may not be lodging firmly in the tension mechanism. Make sure that the thread is firmly settled between the tension discs.
Also keep an eye on what is happening with the thread in the take-up lever (the arm that moves up and down with each stitch on the head of the machine) as the thread may be 'jumping' out thus causing the message.
