SOURCE: 2nd gear does not work on my Yamaha Virago xv250
Sounds like a slightly bent shift fork. The slider gear isn't quite engaging into 2nd. If the bend gets worse then you won't get into first and the pedal will get real stiff in those gears. The repair is best done in the shop.

It sounds like the shifter pawl might not be centered. This is located behind the clutch basket and is a pretty big job to get to, easy adjustment just a pain to get to. You will need a manuel to do it yourself.

Oh wait, are you holding the clutch in and trying to shift down? You have to let out on the clutch after you shift each gear. So the gears can line up with each other to shift.

Sounds more like a shifter pawl problem. I'm not familiar with the RevTech tranny but it should be very similar to the original H-D tranny. The shifter pawl has to be adjusted so that it's got equal travel in both directions. If you're trying to adjust it. Put the tranny in neutral. Now lift the shifter lever until you feel the pawl engage the pin in the shifter drum. Then go back to the middle of the movement range and depress the shift lever until you feel the pawl contact the other pin. The lever should move about the same distance in each direction. If not, the shifter pawl needs to be adjusted. On some bikes, it take some special tools to be able to get to the adjuster. The adjuster is on the left side just in front of the lever that connects to the shifter pawl shaft sticking out of the transmission.

I would have the timing checked. Put new plugs. Sounds like an ignition problem.
