Do you mean the brown flask shaped bottle with the black cap? They can get very tight over time. Best thing is to use a dry cloth or rag & give it your all. Anti Clockwise I'm sure!
If it's very slippy & covered in oil, dirt etc give it a wash with warm water & dish washer fluid. Something that will remove the grease. Dry it off then try again.
The bottle can be removed with the hose on the bottom still attached !!! If you need to hold it in one hand while trying to twist the cap of with your other hand.
If all this fails then may I suggest one of those handy gadgets for opening Jars. They can be picked up in most household goods stores. This should give you the leverage needed.

this model does not come with a dipstick. does require special tool to remove transmission fill plug on the side of the transmission. just fill to the top of the fill hole.

The 4L30E transmission is a "sealed" unit and as such does not have a
dipstick. The only way to check your fluid level is to use the "higher"
of the two drain holes on the bottom of the pan. You *must* do this
while the engine is hot and running, otherwise all the fluid will have
drained to the pan and will come out of the top hole all over you.When
hot, the fluid should come up to the top hole. Stick a finger in, if
you can't feel fluid, use a syphon or hand pump to shoot some up into
it. I would strongly suggest dropping the pan and
replacing the filter if you have over 100k miles. DO NOT have
this trans flushed at a shop as the small galleries are prone to
jamming with the gunk the flush will free up, and will cause the trans
to fail.

The newer power plants do not have a dip stick for checking oil level. Oil level is checked by either the I-drive or by using the BC stalk.
