I have a 2006 Toyota Sienna XLE van, with power sliding doors that quit working with power both at the same time. I suspect a blown fuse, and find that the fusebox is located in a plastic case in teh engine compartment. Do I need to remove the entire plastic case to change the fuse? I would hate to pull the case out and find I have messed up other fuses somehow by doing it incorrectly.

This may or may not work depending on exactly what is wrong with the door, I cant tell what is wrong without looking at it directly however:
You can try starting with all doors closed and unlocked, keys in hand disconnecting the battery and letting the vehicle sit for 2 min or so, then reconnect the battery. Put the key into the ignition and turn it to the on position but do not start the van. Next press the button to disable the power sliding door for the drivers side, then manually open the door fully, wait 5 seconds, then close it manually fully. Press button the button again to re-enable the power sliding door, then try to use it with the automatic feature either by pulling the handle or using your remote, this may or may not work depending on if the door has somehow become de-initialized (it should work now) however if something is broken inside the door closing mechanism or door closed/open sensors, the door will continue to malfunction.

it is harness connection issue. Under driver seat check for any loose connection. make sure all of them connected properly by pressing connector together and it should the problem

Your question is too vague about the "problem". Is it alignment, latching, locking, handle, etc.? Try looking at the following links:

I checked all the fuses. No fuses are blown. The battery is fully charged & the electrical, lights, & radio all work fine. The 2004 Toyota Sienna van still doesn't even turn

Can be a bad contact in one of the cables. A bad contact in a switch or electronic switch. a bad relay, and a dozen other causes.
Please start measuring to where the power comes and where it stops. You are close to the car, we are not. For a correct diagnose use a multimeter and look under the hood.

Check the fuse marked "power outlet" on the fuse panel inside lower left of the dash, there may be more than one marked like this.

Driver side power sliding door in 2006 sienna stoped operating all of sudden, any solution?

Check the owner's manual for a fuse diagram including the high amp fuse box under the hood as most likely the fuse has blown. Lubricate any moving parts on the door as this may have caused the blown fuse.

Look for the power locks (power) fuse in the cabin fuse by the steering column, if that fuse is ok, you may have a body ecu problem.

Both manual sliding doors on my Toyota Sienna will not open

If you can remove the inner door panel you will see a maze of cables running to the various locking points on the sliding door. If the cable going to the FRONT latch on the sliding door is broken (or in my case, simply dislodged inside the door) the door acts as if it is frozen shut.

The front cable is short, and easily replaced even by a newbie mechanic.
