if your are looking for the air conditioner Freon low side port i can only assume that your trying to recharge your ac yourself. DO NOT DO IT!! i work on cars professionally. the only way to properly charge your ac is to evacuate first under vacuum to eliminate all moisture from the system. if you have any moisture in the system and you run it it will cost you thousands of dollars instead of the hundred it would cost to have a professional do it. if the Freon cans worked well mechanics would use them instead of buying thousands of dollars worth of machines to do it.
This is best handled by a shop which specializes in AC repair. I doubt you have the proper equipment to flush, lubricate and recharge the system. It isn't as easy as it once was when you screwed a can of refrigerant to the nipple on the low pressure side of the circuit.
I had the same problem, found out it was the filter which is located in the glove box, open the glove box and the back panel should lift up and out and u will see the white plastic guard for the filter, take it off and pull out the filter and replace it.
I'm not a Lexus pro but having done car stereo installs, common places are under the seats front or rear, or in the trunk well somewhere. May also be in the dash behind the deck.
