Ther is no coil "PACK" on your engine. Each plug has its own coil. Each one is held in place by a single 8mm head bolt that screws into the intake plenum. To change them you simply disconnect the electrical connector, remove the bolt and pull the coil and spark plug boot up out of the spark plug tube. (Reverse proceedure to install.)

SOURCE: 2000 Ford Expedition EB runs rough and begins to shake and stall

I personally would start with the one that runs cylinder #3 and then take it from there.

What would cause a misfire of #1 cylinder on 2000 ford expedition 5.4 liter engine?

these have a problem with coil packs and ignition wires. check the plug it may need replacing, but change all the plugs not just the one. you could have a dirty or a bad injector for that cylinder or even a vacuum leak at the intake for that cylinder. something else is you could have poor compression on that cylinder.

I have a 2000 Ford Expedition with 157,000 miles. Do I need to be concerned about replacing the timing chain?

no you should not worry about changing timing. unless its not starting or something is really wrong that points you to the timing chain. i have 250,000 on my f150 never had no problems with timing chain. maybe a coil pack or sparkplug but no timing chain.
