Do you mean the timing marks on the timing belt or the cam? If it's the belt, there should be 3 lines on the belt. Put the belt on the cams and turn them until all 3 of them line up with the timing marks on the cams. If it is a timing issue, you need a timing light to set the timing. There should be a pointer or timing mark on the cam with timing marks on it. You need to align the mark on the cam with the pointer while the engine is running with a timing light.

but spark or cam?
all cars have cam timing and spark timing.
some newer cars the cam sensor is fixed, but not yours. i bet.
why not , pray tell, say what you removed. or fiddled with.

This is mainly a matter of having the timing marks lined up along the timing belt at top dead center, and then checking that the spark advance is working OK . See here p30 and 44
The crankshaft gear has a center punch mark on gear and the intermediate gear has a line mark on gear. Rotate the crank until the #1 piston is top dead center. In doing so the crankshaft mark should line up with the intermediate gear mark. (crankshaft mark at 1 oclock and intermediate gear mark at 7 oclock). At the same time look at the cam gear and the small hole should be at the top and the two larger holes should align with the bearing cap. ((parting line should be looking at it will be 50/50 when you look at the front of cam gear). Remember to remove belt before timing engine correctly. Also make sure the negative wire is disconnected from battery before starting this job. Hope this will help you.
all marks align at noon,both lines on camshafts point up ,don't follow any marks on face of crankshaft gear follow line located on reluctor this is part of the crankshaft gear on back of it, the line on the reluctor needs to face noon and match a line on the oilpump where the crankshaft position sensor is installed good luck

Get a T belt from toyota. It has 3 white lines on
it and and arrow mark. 2 lines are solid and one is broken. Put the
arrow facing away from the engine. The cam gears have a dot punched
into them near a certain tooth. This is where the solid white lines
will line up with. The crank gear will have a dot also. It will line up
at 90 degrees from the top, towards the front of the car. This is where
the broken line will match up with. With the belt installed with all
lines on their proper dots and arrow away from the engine, you valve
timming is correct. Of course you need to remove and compress the tensioner also. I assume you know this already since you didn't ask.
