I have the 4 speed auto box with overdrive with the 4.0 V8. In drive it suddenly will not take off from a standing start in 1st but 4th. I have to drive it manually and then i only have 4 gears and when i change manually to 2/3, 2/3 are on top of each other and when changing to D4 4th takes a very long time to engage and no overdrive. Can you help?

There isn't anything wrong with your car. This is perfectly normal. I have a 2000 ES300 as well as other people that I know and it operates in the same way. Overdrive doesn't mean the same thing for every car or transmission. In our case overdrive is simply a reference to the top gear which is 4th gear. When you switch overdrive off(which is depressing the button causing the O/D OFF dash light to illuminate,) the highest gear becomes 3rd because you turned 4th gear or overdrive off. The application scenario for this is usually mountainous terrain where a lot of uphill driving makes it more feasible not to up shift into 4th gear while ascending and descending. It will not switch back to overdrive or 4th gear until you re-engage it by turning the overdrive back on or repressing the button. Again, when the O/D OFF dash light is illuminated, overdrive which is 4th gear is off and the highest gear will then become 3rd. It will never shift into 4th gear while this light is illuminated or overdrive is off. When you press the button and the O/D OFF dash light is NOT illuminated overdrive is on and therefore it will shift into 4th gear as usual. This applies to all 97-2001 ES300's and probably any other car you drive tat exhibits this same type of change when overdrive is turned off.

It sounds like you need to get the auto flashed(computer re programming) I Live in Australia and thats whats happening here,the cars are being returned to the dealer to re programme the trans to correct the shifting faults,you would ask your self why is the oil being replaced or topped up on a new car and Lexus is known for its quality control,and technicians training,you may want to go to a different dealer if youre not getting satisfaction,this will fix your issues

Start by checking the least expensive items: Make sure you are not activating the O/D switch in the shift stick inadvertently. Look at the display panel, if the "Eng" light is on, you must get the code and repair the affected item. If you have knock sensors and/or oxygen sensors malfunctions that may prevent the transmission from shifting to O/D. Check the ground cables located in the engine compartment, especially the ones behind the air intake manifold. If they are loose or broken it affects the ECU (car computer) and causes the transmission to work erratically. If they become disconnected it may burn the ECU and shut down the car. Now going to more profound mechanics, it could be a sticking shift solenoid valve, your RX300 transmission have five of those located were the transmission oil strainer (filter) is. It can also be an ECU malfunction and/or related to the wiring from the ECU to the transmission connectors. Always start from the least expensive and recheck the obvious. Good luck.
