I have a 2008 Toyota Highlander. If I lock the doors remotely and then go around to the trunk and pull the handle twice, all the doors (including the trunk) unlock and the interior lights illuminate. So basically, anyone can get into my locked car as long as they try the trunk twice.

There's a button on the glove box toward the center console, try turning it off and see what happens.

SOURCE: 1998 Jetta Passenger Door Won't Unlock

I had a similar problem with my son's 1998 Jetta K2 4 door. I sprayed loads of WD40 into the locking area from the end of the door through the clip slot, worked the mechanism sprayed some more and finally it start working. My mechanic said these are very prone to moisture causing rusting on the moving parts.

SOURCE: remote starter does not work but will unlock and lock doors.

You need to first press the LOCK button and then immediately hold down the START button for 2 seconds - voila!

Rear PAX door will not open inside or out, when unlocked with inside door lock or with remote.

The cable connecting the handles to the latch mechanism has probably come loose or has broken. You will have to open up the door to see. There are usually only 2 or three screws holding the door on. One behind the release handle and one or two behind the armrest/door pull. If you can't tell how the cable should be connected compare to the one on the working side.

My highlander 2008, the doors can be locked and open with key and remote but cannot open the trunk with key and remote

To make the rear door open, you have to hold the button for a while. If the battery is weak, that will drain the power and the door will not open. Try the other remote and see if it works. At that point, I would replace the batteries in both remotes anyway.
That would be the fastest and cheapest way to eliminate one possible problem.
If the inside open button works to open the door, I would say the mechanism is working fine and it is the batteries.
I had to replace the batteries in both my remotes at the 3 year mark. That should be about now, the second time, for yours.

We have a backpack strap caught in the back hatch lock on our Highlander and now cannot open the trunk- any suggestions?

you will have to have some 1 operate the lock manually and some one to operate the hatch the person operating the hatch will have to pull very hard or may even need to pry it open while pulling on the door handle put something over the bumper so as not to scratch it and use a tire iron or something along those lines it will take a little effort but you can do it.

Back hatch door closed on strap of backpack-now can't open door

Try pressing down on the trunk /hatch and at the same time turn the key to try and open it. It might take a lot of downware pressure to finally get it to come open.
