I had similar on 02 Highlander. Source was leak from around window trim (rust) and roof racks connector entering and going down the windshield post behind the trim. Gently remove the interior plastic trim piece to the right of the windshield to see any water coming down there. Find source, disassemble whatever is required and use good sealant like supra expert.

2011 Toyota Highlander. Had no luck finding the reset switch, HOWEVER, the warning indicator also includes the SPARE tire (there is a BT sensor in the valve stem which operators on a tiny battery which signals to the car as well). We had filled the 4 vehicle tires properly, and was still showing the tire pressure warning. Researched it and saw the SPARE is include with this monitoring system for the vehicle. Checked and filled the spare, the warning indicator turned off.

That sound can indicate a problem with the VVT phaser gear. Do you use the correct weight oil and change it no more than every 6,000 miles?

May be bad sensor.

Reset the Tire pressure monitoring system light. The reset button is located under the steering side under the finish panel. You have to bend down to see the button. Operate the tire pressure warning system reset switch only after the inflation pressures of all 4 tires have been adjusted on the vehicle. To initialize the system, press and hold the tire pressure warning system reset switch for 3 seconds or longer with the power switch turned IG-ON

did you recently change to larger tires? could be a possible cause.
